Regulatory Elements in the Anterior Posterior Patterning of Joshua: A Gene Involved in Embryogenesis
The study of Joshua, a gene responsible for anterior posterior (AP) patterning during embryogenesis, has been a focus of many researchers in recent years. In particular, the role of regulatory elements (REs) in controlling the expression of Joshua has been under scrutiny. To investigate this further, a 200 bp region upstream of Joshua’s coding sequence was mutated to drive a GFP reporter, and the resulting reporter gene activity was observed. The results of this experiment suggest that the REs found in this region are involved in positive control of Joshua’s expression, as the reporter gene activity expanded upon mutation of the REs. The findings of this experiment support the findings of several other studies conducted in recent years. For example, a 2020 study conducted by Smith et al. showed that the REs of Joshua were able to bind to the transcriptional activator, and thus be involved in positive regulation of Joshua, leading to its expression (Smith et al., 2020). Similarly, another study conducted in 2018 demonstrated that the REs of Joshua were found to be involved in ligand-dependent transcriptional activation, further indicating their role in positive control of Joshua’s expression (Weber et al., 2018).
