Phenotypic Analysis of E. coli trp Operon Mutants with Deletion of Adenosine at Position 35 of the Leader RNA

The expression of the trp operon enzymes of Escherichia coli is regulated by the leader RNA of the operon. The leader RNA contains a sequence of 35 nucleotides, of which the 35th nucleotide is adenosine (A) (Wang, 2016). The deletion of A at position 35 of the trp operon leader RNA results in structural changes in the leader RNA, leading to the production of altered mRNA molecules (Chu et al., 2018). The altered mRNA molecules lead to the expression of the trp operon enzymes in an altered fashion. This altered expression of the enzymes is expected to lead to a phenotypic change in the E. coli strain. Previous studies have reported that the deletion of A at position 35 of the trp operon leader RNA leads to an increase in the expression of the trp operon enzymes (Hsu et al., 2017). This increased expression of the trp operon enzymes leads to an increase in the production of tryptophan and its derivatives in the E. coli strain. This increased production of tryptophan and its derivatives is expected to lead to a phenotype of increased tolerance to tryptophan starvation in the E. coli strain (Hsu et al., 2020). Cont…
