The Challenges of Induced Pluripotent Stem Cell (iPSC) Reprogramming

The process of induced pluripotent stem cell (iPSC) reprogramming can be a complex and difficult one. Despite its promise in regenerative medicine and tissue engineering, the challenges of the process must be addressed before the potential of iPSC reprogramming can be fully realized. These challenges include the generation of a sufficient number of viable iPSC, the establishment of stable and uniform iPSC lines, and ensuring that the iPSC are safe and effective for clinical use. In terms of generating a sufficient number of viable iPSC, there is a need to understand the reprogramming kinetics and optimize the culture conditions to enhance reprogramming efficiency. Studies have shown that the reprogramming process can be improved by increasing the expression of certain transcription factors (Liu et al., 2016; Shi et al., 2021).
Additionally, the establishment of stable and uniform iPSC lines is a critical component of iPSC reprogramming. It is important to ensure that the iPSC are morphologically and functionally similar to embryonic stem cells, and that the iPSC do not form teratomas or other forms of tumors (Araki et al., 2020; Zhang et al., 2021).
