The Role of Enzymes in Connecting the Pentose Phosphate Pathway to Glycolysis
The pentose phosphate pathway (PPP) and glycolysis are two interconnected metabolic pathways that are critical to the production of energy in the cell. The two pathways are linked by two enzymes: glucose-6-phosphate dehydrogenase (G6PD) and 6-phosphogluconate dehydrogenase (6PGD). G6PD catalyzes the oxidation of glucose-6-phosphate to 6-phosphoglucono-?-lactone and hydrogen peroxide, while 6PGD catalyzes the oxidation of 6-phosphoglucono-?-lactone to 6-phosphogluconate and NADH (Mollah et al., 2016). The PPP generates NADPH, which is used to reduce ribulose-5-phosphate to ribose-5-phosphate and is then used for the synthesis of pentose sugars. The NADPH is also used to reduce NAD+ to NADH, which is then used in glycolysis to produce ATP (Rajput et al., 2021). This conversion of NAD+ to NADH is essential for glucose metabolism and is known as the oxidative phase of the PPP. G6PD and 6PGD are both essential enzymes in linking the PPP and glycolysis. G6PD is responsible for the oxidation of glucose-6-phosphate to 6-phosphogluconoate. Cont…
